On your way to visit Zelda for the second time in the game shortly after completing Lakebed Temple you'll come across a man named Jovani. You kind of stumble into his house by accident while in Wolf Form and you get introduced to one of the main collectibles in the game - Poes.
Poe #38 - Located in Gerudo Desert
The first Poe for us to grab in Gerudo Desert is located right by where you land if you take Fyer's Ride in Lake Hylia <Name check Fyer make sure right>. He's just a bit to the south, you'll be able to see him if it's night out. If you didn't grab the Warp Portal your first time in Gerudo Desert the only way back will be taking the Lake Hylia cannon
Poe #39 - Located in Gerudo Desert
The second Poe for us to grab in Gerudo Desert is by the Warp portal; also the Cave of Ordeals. You need to Clawshot up to this platform at night which is where you'll find the Poe and be able to open up the Warp Portal. I've included a map location for this Poe as well since he's nearby such an important land mark.
Poe #40, #41 and #42 - Located in Gerudo Desert
In northern Gerudo Desert, the very wide open flat part of the desert where you had to fight the Bulbins on the towers earlier in the game you'll find 3 more Poes. They are located on a raised piece of land which is found on the cusp of southern and northern Gerudo Desert. Take a look at my screen shots for the exact locations.
You'll need to Clawshot onto a tree to get up here, much like you had to do to reach the Warp Portal. There will be a single Poe flying around here at night which you'll want to kill - then you'll want to turn into a wolf and hit your senses. You'll notice that there's a spot for you to dig in the middle of the three skulls (as seen in my screen shot).
This will drop you down into a cave with two more Poes for you kill. If you'd like there's also an Orange Rupee you can grab while you're here - light all the torches in this cave to make the chest appear.
Poe #43 - Located in Gerudo Desert
This Poe can be found just before the zone line leading to the Bulbin fortress. If you've grabbed all of the Howling Stones up until this point in the game, you met a Golden Wolf very close to this location as well. The Poe is found tucked away in the corner area, a little bit past where you found the Golden Wolf.
Poe #44 - Located in Gerudo Desert
You'll find this Poe in the burned down building where you fought the big fat King Bulbin just before entering Arbiter's Grounds.
Poe #45 - Located in Gerudo Desert
This Poe is found just outside the entrance to Arbiter's Grounds. Can't miss him!
Poe #46 - Located in Arbiter's Grounds
You'll find this Poe as well as 3 others by normally progressing through Arbiter's Grounds. Since they are part of the game's story and required to advance the game, much like the first Poe we captured, i'm not going to go into a lot of detail about them.
The first one is captured in the main room and opens you up to this event.
Poe #47 - Located in Arbiter's Grounds
You'll find this Poe as well as 3 others by normally progressing through Arbiter's Grounds. Since they are part of the game's story and required to advance the game, much like the first Poe we captured, i'm not going to go into a lot of detail about them.
Poe #48 - Located in Arbiter's Grounds

Poe #49 - Located in Arbiter's Grounds
You'll find this Poe as well as 3 others by normally progressing through Arbiter's Grounds. Since they are part of the game's story and required to advance the game, much like the first Poe we captured, i'm not going to go into a lot of detail about them.
